Alcohol alters brain structure at the molecular level

Specific proteins within the brain's building blocks, the neurons, are damaged by alcohol.

Joint Spanish/English research has identified, for the first time, the structural damage caused in brain cells by excessive alcohol use. The research was published in the April 2014 biomedical sciences journal, PLOS One. This breakthrough by the University of the Basque Country and the University of Nottingham opens up pathways for generating new medications or therapies to enhance the life of recovering alcoholics.
Binge drinkers and those with the disease of alcoholism commonly experience learning and memory deficits, impairment of decision making and poor motor skills. They sometimes show behavioral changes, as well, plus anxiety and depression from cumulative effects of intoxication and alcohol withdrawal. Excessive alcohol consumption can result in a reduction of brain weight as well. The new discovery identifies, for the first time, the damage to the brain from alcohol goes down to the cellular level.
The research team determined alcohol causes alterations in the proteins in neurons of the prefrontal zone of the brain. Neurons are the basic building block of the nervous system... the prefrontal zone is the most advanced brain region in terms of evolution and controls functions such as planning, designing strategies, working memory, selective attention or control of behavior.
In the research, doctors studied the postmortem brains of 20 men and women diagnosed with alcohol abuse/dependence, alongside another 20 non-alcoholic brains. Studying the prefrontal cortex, researchers detected alterations in the molecular “cytoskeleton” in the brains of alcoholic patients. These alcohol-related changes in the brain structure can affect the capacity for making connections and the functioning of the brain – and could largely explain alterations in thinking and learning – attributed to persons suffering from alcoholism.
The researchers aim to further identify these molecular changes in order to be able to, on the one hand, link them with the processes of alcohol abuse and dependence and, on the other, generate new pharmaceutical drugs and therapeutic options that reverse the alterations produced by alcohol, enhancing the quality of life of alcoholic persons and reducing the mortality rate due to alcoholism.

Study finds counterintuitive father/son alcoholism link

A son's susceptibility for an alcohol use disorder may actually drop if dad drinks abusively before conception according to University of Pittsburgh School of Medicine rodent research. The June 4 study results suggest male offspring might drink less.
The research finds mice chronically exposed to alcohol before breeding had male offspring that were less likely to consume alcohol and were more sensitive to its effects. This could be a fresh take on the prior human studies indicating that alcoholism can run in families, particularly father to son. Research student, Andrey Finegersh, noted, "We suspected that the offspring of alcohol exposed sires would have an enhanced taste for alcohol, which seems to be the pattern for humans. Whether the unexpected reduction in alcohol drinking that was observed is due to differences between species or the specific drinking model that was tested is unclear."
Lead researcher, Gregg E. Homanics, notes, "Rather than mutation of the genetic sequence, environmental factors might lead to changes that modify the activity of a gene, which is called epigenetics. Our mouse study shows that it is possible for alcohol to modify the dad's otherwise normal genes and influence consumption in his sons, but surprisingly not his daughters."
The researchers plan to examine other drinking models such as binge drinking, identify how alcohol modifies the genes, and explore why female offspring appear unaffected.
In the National Institute on Alcohol Abuse and Alcoholism (NIAAA) and National Institutes of Health (NIH) funded study, male mice were exposed to ethanol vapor over five weeks, leading to blood alcohol concentrations (BAC) slightly higher than the .08 legal limit for U.S. drivers. The exposed mice were mated to females who had not been exposed to alcohol. Compared to those of ethanol-free fathers, adult male offspring of ethanol-exposed mice consumed less alcohol when it was made available and were less likely to choose to drink it over water. Also, they were more susceptible to drinking's affect on motor control and an alcohol-induced reduction of anxiety.
The UPMC study, while contradictory to human gene studies, does confirm that alcohol consumed and its acetaldehyde byproduct leave a biological imprint on your DNA. The idea that relapse is part of recovery is only partially true.  Nine of 10 ex-drinkers go back to alcohol... true.  Some don't make it back to sobriety.  And if the stats hold true, relapse is NOT part of the recovery of the one in ten who defied the odds.  Relapse can be part of recovery, but doesn't have to be, nor should it be a justification for letting sobriety lose its priority.

Classifying alcohol as "not a drug" like cocaine or opiates is inaccurate.  Alcohol is the second-leading drug of abuse behind caffeine.  Only public perception (or mass denial) says it isn't a drug and that alcoholism isn't a drug addiction.
